Pharmajor is a European pharmaceutical company developing CONSILUM, a novel extended-release formulation of silodosin for use as a reversible, non-hormonal, once-daily oral male contraceptive. Silodosin is an FDA-approved alpha-1A adrenergic receptor (α1A-AR) antagonist with a well-established clinical safety record in wide chronic use in men worldwide. Pharmajor is directing a late-preclinical development effort that leverages silodosin's existing clinical data package to pursue a streamlined regulatory pathway for male contraceptive approval. Silodosin works by selectively relaxing smooth muscle in the male reproductive tract, an effect that reliably suppresses the emission of sperm and semen during ejaculation without affecting hormone levels or sperm production. Clinical studies in men have demonstrated that this effect is reversible upon discontinuation and produces no alteration of sperm integrity. Pharmajor has developed CONSILUM, an extended-release oral formulation of silodosin designed for once-daily dosing and continuous contraceptive efficacy. Our Grant
MCI's grant to Pharmajor supports nonclinical activities required to move into later stages of the clinical trials approval process including Phase II efficacy studies. Because silodosin's safety in humans is well-documented through decades of clinical use, CONSILUM can leverage a 505(b)(2) regulatory strategy using existing clinical data as part of the approval package, potentially compressing the development timeline relative to new chemical entities. |
